腦中風一向是普及全球的導致死亡或成年人殘障之重要導因,依其病理機轉可分為「出血性中風」與「缺血性中風」兩大類,其中缺血型中風就佔了所有腦中風患者的七至八成。目前臨床急性處理上主要是以血栓溶解劑rt-PA來治療,然而其限制甚多,除了必須在急救黃金時間(三小時)內注射,還要排除具腦出血可能性的患者。因此,開發出更合適的預防或治療中風之藥物是許多學者的目標。本研究目的乃藉由評估細胞的黏著表現,探討川芎嗪(2,3,5,6-tetramethylpyrazine, TMP)對氧糖剝奪損傷的PC12細胞之保護作用。
細胞缺氧合併培養基缺糖可以造成擬體內缺血時的變化,本研究以Na2S2O4(5 mM, 1 h)去氧配合無糖的磷酸鹽緩衝液(Phosphate-Buffered Saline, PBS)置備出擬體內缺血模型,實驗結果顯示,PC12細胞的黏著剪力強度顯著地降低,顯示此種傷害模型的確造成了PC12細胞的損傷。而經由兩種劑量(50 μg/ml, 500 μg/ml)的TMP進行藥物預處理及後處理,可使受到擬缺血損傷之PC12細胞的黏著剪力強度顯著地提高,而且成劑量依賴(dose-dependent)的效果,顯示了TMP對於擬缺血損傷的PC12細胞具有保護與修復的作用。經由與相關文獻的相互印證,本研究證明了以比較細胞黏著剪力強度大小來判斷細胞活性的方式,不失為一可行的簡易藥效評估指標。
Cerebral stroke is a leading cause of death and adult disability worldwide. According to the pathological mechanisms, strokes are classified into two major types, hemorrhagic and ischemic strokes. Ischemic stroke accounts for 70-80% of all strokes. At present, the Intravenous rt-PA is mainly used for treatment of acute ischemic stroke . However, it has a variety of restrictions and is suggested to give the patient an injection within the golden hours (three hours) with exclusion of the probably patients of cerebral hemorrhage. It is a challengeto develop proper medicines for stroke prevention and treament. Therefore, the purpose of this study was to investigate the effect of 2,3,5,6-Tetramethylpyrazine (TMP), one of the most important active ingredients of traditional Chinese medicine Ligusticum wallichii Franchat (Chuan Xiong), on PC12 cells after oxygen-glucose deprivation in terms of quantatative cell adhesion forces measured by the cytodetachment technique.
Cells were cultured in a hypoxia and glucose-free medium to simulate ischemia. We use sodium dithionite (5 mM, 1hr) to deoxygenate glucose-free PBS to establish an ischemia-like model. The results indicate that the adhesion shear strength of PC12 cells in the OGD group significantly lower than that in the control group. It reveals that this model indeed caused PC12 cell damage. After pre-treated and post-treated with TMP, 50 μg/ml and 500 μg/ml, respectively, the adhesion shear strength, adhesion force per unit area, of PC12 cells increased significantly, but was dose-dependently. It reveals that the TMP can protect and repair PC12 cell damage caused by the ischemia-like model. As compared with previous studies, our study indicates that the evaluation of the cell activity by the cell adhesion shear strength may be a useful drug assessment index.
