巨細胞病毒屬 (Megalocytivirus) 是虹彩病毒科 (Iridovidae) 之第五種被發現的病毒。巨細胞病毒是雙股DNA病毒。自 1990 年代初期發現以來,它已經引起重大疾病和經濟損失。其中,傳染性脾臟及腎臟壞死病毒 (Infectious Spleen and Kidney Necrosis Virus, ISKNV) 在許多亞洲國家造成了對水產養殖業的巨大損失。致病性微生物會誘導宿主改變代謝途徑讓自己利於生存,並且具有許多調節宿主免疫的機制,以促進本身的感染和傳播效率。這些病原體誘導之代謝產物可以反映病原體調節之免疫反應過程,並且可以在研究後推導其途徑。在這項研究中,以超高效能液相層析串聯式質譜儀 (UHPLC-MS/MS) 分析傳染性脾臟及腎臟壞死病毒感染石斑魚肝臟上皮細胞株後造成之代謝物變化,並且就固醇類荷爾蒙生物合成代謝途徑來探討病毒感染之分子機制。針對代謝途徑中的雌三醇 (Estriol) 及 孕酮 (Progesterone) 分子也發現了初步抑制ISKNV的效果,之後的研究將要探討代謝物在病毒感染中所扮演之角色,找到能抑制病毒傳播的條件,以實現水產養殖中 ISKNV 之預防及控制。
Megalocytivirus is the fifth discovered species of the iridovidae family. It is a double-stranded DNA virus. Since its discovery in the early 1990s, it has caused significant disease and economic loss. The widely spread aquatic virus, infectious spleen and kidney necrosis virus (ISKNV), has caused severe disasters to the aquaculture industry in many Asian countries.
Pathogenic microorganisms induce host metabolism and get profits to survive. Host-adapted pathogens have many mechanisms to modulate host immunity to facilitate infection and transmission. These pathogen-induced metabolites can reflect pathogen-regulated immune response processes, and their pathways can be deduced after study. In this study, ultra-high performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS) was used to analyze metabolite changes caused by ISKNV infection of grouper liver epithelial cell lines and to explore the molecular mechanism of viral infection based on the steroid hormone biosynthesis pathways. Estriol and Progesterone in the metabolic pathway have also been found to have a preliminary inhibitory effect on ISKNV. Subsequent research will explore the role of metabolites in viral infection and find compounds that can inhibit the viral replication conditions to achieve prevention and control of ISKNV in aquaculture.
