Micorarray是一門新興且具有前景的生物科技,它可以同時監督數千個基因的表現。在實驗的過程中,有許多系統性的變異會影響所測得的基因表現值;而袪除這些變異的過程就叫做normalization。Dudoit et al. (2002)建議使用lowess來進行normalization。在本文,我們指出Dudoit et al. (2002)之lowess normalization的缺點,進而從一個新穎的觀點提出在cDNA microarry實驗下的normalization方式。我們以模擬的方式來證明所提的方法優於lowess,並以成功大學微生物免疫所提供的微陣列資料來說明所建議方法的應用。
Microarray is a new and promising biotechnology which allows people to monitor the expression levels for thousands of genes simultaneously. There are many sources of systematic variation which may affect the measured gene expression levels during the fabrication of microarray. The process of removing such variation is called normalization and Dudoit et al. (2002) proposed lowess method to normalize the microarray data. In the thesis we suggest a new normalization procedure for cDNA microarray experiment. Statistical simulations show that our proposed method is superior to lowess normalization. The proposed approach is illustrated with gene expression data provided by College of Medicine, NCKU.
