臺灣龍膽石斑魚(鞍帶石斑魚)為一產量高且經濟價值高之養殖魚類,由於近年來養殖技術的成長,擁有穩定之產量,故本實驗探討石斑魚受精卵萃出物之分子量分布、生化活性,進而研究發展為高附加價值之美容保養品。
石斑魚受精卵經處理後,以不同比例鳳梨酵素與木瓜酵素共同水解五小時與十小時,再以FPLC測定發現鳳梨酵素及木瓜酵素以4:1 (w/w) 1 g加入蛋白質濃度1.4 µg/µl之100 ml魚卵萃取液水解十小時後,分子量鑑測得6512 Da以上大分子量水解效果較佳。再進行抗氧化測試,包含清除DPPH自由基能力、螯合亞鐵離子能力、還原力測定、總酚含量測試。經比較發現未經水解之魚卵萃出物效果反而較佳。以超濾膜將未水解魚卵萃出物區分為不同分子量範圍,並以FPLC鑑測分子量分離功效,再將分離出不同分子量之萃出物做一系列抗氧化測試,實驗結果以1 kDa以下萃出物較佳。
在細胞培養上,使用HEPM cell及HK-2 cell做MTT assay,測試細胞增殖率,實驗結果發現未經水解之萃出物比水解後之萃出物佳。此外受精卵萃出物在重金屬及微生物檢測報告中均未檢出,顯示此成分無汙染。進而開發為以3 %未水解萃出物為基底之保養品,在臨床肌膚測試一週後,保濕效果達68.3 %且淡斑效果達31.8 %,具有成為保養品之潛力。
Taiwan Epinephelus Lanceolatus is a highly economic fish species. The cultivated technique for Taiwan Epinephelus Lanceolatus has been improved and can maintain a steady production for the rising market. This fish contains proteins and bioactive ingredients, which can increase economic benefits. In this study we used FPLC to measure the molecule weight of hydrolyzed zygote extractions and found that the molecular weight over 6500 Da were completely hydrolyzed by papain enzyme and bromelain after hydrolysis for 10 hours. The bioactivity of radical scavenging activity, ferrous ion chelating ability, total phenolic content, and ferric reducing antioxidant power for the zygote extractions were assayed. The result showed that non hydrolyzed zygote extraction exhibits higher antioxidant activity than hydrolyzed. Thus, the non hydrolyzed zygote extraction was separated into different molecular weight. And found that molecular weight less than 1 kDa showed the highest antioxidant activity. The non hydrolyzed extraction also showed possessing the ability to boost the cell growth. Besides, this extracted did not contained heavy metal and microbial. A cosmetic consisted of 3% non hydrolyzed zygote extraction and 97% basis medium were tested by applying to a group of 30 people with age between 30-40 years old, and found that the face freekles, pores, and moisture retention improved after two week’s apply. This result indicated that this species has the potential to develop into cosmetic raw material.
