(pyrazole)之衍生物已有文獻報導指出,具有生理活性和藥理作用,例如:抗癌、止痛劑、退熱劑...等效用。還有生化方面的應用,如:血小板抗化劑、除草劑….。除此之外,近年來此類具Pyrazole結構之化合物常被用來作為有機發光二極體 (OLED和PLED)。
N3-苯甲基-N5-環己基-1-(4-乙氧苯基)-1氫-pyrazole-3,5-二甲醯胺等pyrazole之衍生物已經被證實具有抑制幽門螺旋桿菌(H. pylori )Dihydroorotate dehydrogenase (DHODase)的藥理療效。
本論文中實驗之第一部份是利用本實驗室開發已久的雪梨酮化合物經由1,3-偶極環化加成反應,隨後再經過醯胺化反應以及Ritter反應,即可得到具有抑制幽門螺旋桿菌DHODase酵素的衍生物。而此新穎的實驗方法比文獻所呈現的方法,要來得簡單和迅速的獲得產物。可是在位向選擇性和產率方面卻不理想,而造成整體的產率偏低,所以實驗第二部份就是設計改善其選擇性和產率的研究,於本論文中有詳細的描述。
Derivatives of pyrazole have biological activities and pharmacological actions had been reported by many researchers, such as : Anti-cancer, analgesics, antipyretic...etc. Many molecules containing pyrazole were also used in organic light-emitting diode in recent years.
Pyrazole analogues were demonstrated to be potent inhibitors of H. Pylori DHODase. These compounds are competitive with coenzyme Q6 and were highly selective for H. pylori over human DHODase. In our work, a new efficient synthesis method of pyrazole (5) was developed via 1,3-dipolar cycloaddition of sydnone with acetylene, Ritter reaction and amination.The chemoslectivity of sydnone derivatives with acetylene would be studied to improve the desired yield in 1,3-dipalor cycloaddition.
