本研究探討台灣鯛、虱目魚以及燦麗魚魚鱗膠原蛋白膜交聯之生醫材料製備方式,以及其膠原蛋白膜特性分析。主要選用幾丁聚醣、梔子素、核醣,在不同濃度下與各魚種的魚鱗膠原蛋白進行交聯,冷凍乾燥成膜。於鑑定膠原蛋白方面,使用了Bradford Protein–Binding assay、SDS-PAGE以及傅立葉紅外線光譜分析儀。而在測定交聯後膠原蛋白膜的特性上,使用了熱差式掃描分析、掃描式電子顯微鏡、寧海德林自由胺基含量測定對其做分析。Bradford Protein–Binding assay得到的結果顯示凍乾完的產物均約50%左右為膠原蛋白,SDS-PAGE確認均為TypeⅠ型膠原蛋白,傅立葉紅外線光譜測得amideΙ(1650cm-1)、 amideⅡ(1550cm-1)、amideⅢ(1235cm-1)的三個膠原蛋白訊號。DSC得到的Td值證明幾丁聚醣、核醣對台灣鯛、虱目魚以及燦麗魚的魚鱗膠原蛋白有較好的交聯作用,使熱變性溫度小幅度上升;而梔子素僅對台灣鯛的魚鱗膠原蛋白有較好的交聯現象,於其他兩種魚種則否,反倒有降低熱變性溫度的現象發生。由SEM中觀察到的結果也符合上述現象,交聯較好的薄膜組織緻密少孔洞,而與梔子素交聯的虱目魚與燦麗魚魚鱗膠原蛋白膜則組織疏鬆孔洞多,薄膜本體也較為破碎。寧海德林自由胺基含量測定的結果也接近上述現象,虱目魚、燦麗魚魚鱗膠原蛋白與幾丁聚醣、核醣交聯後的薄膜,其自由胺基含量較低;與梔子素交聯則較高,台灣鯛魚鱗膠原蛋白薄膜則看不出明顯差異。
最後將各魚種物理性質較好的薄膜進行醫用敷料的活體試驗,以小白鼠做為實驗體,結果顯示台灣鯛與虱目魚的交聯後膠原蛋白膜均有幫助傷口癒合的能力,而燦麗魚的則較無明顯的效果,故可以驗證本實驗製成的魚鱗膠原蛋白膜在醫用敷料的應用上有良好的潛力。
In this study, we investigate the method for preparing and charactering the biomaterial film by cross-linking Taiwan Tilapia, Milkfish and Petenia splendida scale collagen with chitosan, genipin and ribose. First, we will characterized the collagen by using Bradford Protein–Binding assay, SDS-PAGE and Fourier Transform Infrared Spectroscopy (FT-IR). After cross-linking with the cross-linking agent, we dry the sample with Freeze Dryer System. Then the cross-linking collagen film was characterized by using Differential Scanning Calorimetry(DSC), Ninhydrin assay and Scanning ElectronicMicroscope(SEM). Finally, we choose the best physical property film from each fish and conduct an in vivo test on rate.
