在先前研究指出,IgT 為特異屬於硬骨魚類的免疫球蛋白,為主要的黏膜抗體影響黏膜層的免疫反應,且黏膜免疫反應亦為抵禦病原體的第一道防線,但其功能仍未了解透徹。而在點帶石斑魚中,IgT 和黏膜免疫的功能仍也有待探討,因此本研究將探討點帶石斑魚IgT (Orangespotted
grouper immunoglobulin T,osgIgT) 其分子選殖與特性分析。利用
即時聚合酶鏈式反應偵測,osgIgT 廣泛表現在免疫組織與黏膜相關淋巴
組織中,如:頭腎、脾臟、鰓與小腸,然而其蛋白表現主要位於黏膜相關
淋巴組織中。在LPS、Poly (I:C)與寄生蟲刺激處理下,osgIgT 基因表現一會呈現不同的趨勢差異性。另一方面,探討在NNV 病毒感染處理下,osgIgT 基因於黏膜相關淋巴組織中會隨病毒複製數同之上升兩倍,並隨感染天數而異,呈現差異性變化。進一步探究osgIgT 與NNV 病毒的相互作用關係,以免疫螢光染色觀察,點帶石斑魚IgT 在小腸與鰓亦有與NNV 病毒共定位的現象。綜合以上結果,osgIgT 在不同免疫刺激中可能具差異性調節達到免疫作用,且參與黏膜層上抵禦病原體的重要角色。
Previous reports had demonstrated that IgT, the antibody considered as specific to teleost fish, is the main immunoglobulin induced in the mucosa on pathogenic challenge and influences on mucosal immune regulation, and mucosa immunity is also the first line of defense against pathogens. However, its function is not being known well thoroughly. In orange-spotted groupers, the function of IgT and mucosal immunity are also known less so far. As a result, this study demonstrated the molecular cloning and characterization of Orange-spotted grouper immunoglobulin T (osgIgT). From the qPCR analysis, osgIgT widely expressed in immune tissues and mucosa-associated lymphoid tissues (MALTs), such as head kidney, spleen, gills and intestine, while osgIgT merely expressed in MALTs in
protein level. Under LPS, Poly (I:C) and parasites challenge, it showed differential expression of osgIgT in response to immune stimulations. On the other hand, during NNVinfected treatment, the level of osgIgT increased 2-fold following as virus copy number in MALTs, and appeared different expression levels to response NNV. Further, the relationship of osgIgT and NNV was investigated by immunofluorescence. The results showed that osgIgT and NNV colocalized together in infected intestine and gills. Taken together, these present results have implicated the effect of osgIgT through differentially regulated to
achieve immune function and pointed out its possible involvement in mucosal immune responses of in orange-spotted groupers.
