近年來由於螢光技術的普及,加上使用時的簡單方便以及訊號的立即回饋性,分子型感測器已成為當今熱門的研究題材。而在生化領域中,綠色螢光蛋白由於能在生物體中透過分子間氫鍵而產生剛性結構,以至於在該環境下的螢光量子產率可高達0.8,並且在過去二十年來被許多研究人員廣泛地用作追蹤生體分子的螢光標籤。但是,若將其中的發光團(p-HBDI)單獨分離出來,其在激發態時會藉由環外雙鍵的扭轉而進行光異構化反應,導致在一般溶劑中螢光量子產率僅剩下10-3至10-4。本實驗將EDTA的性質結合至綠色螢光蛋白,合成出兩種皆對銅離子具有選擇性的多牙配位基,o-DOIMAA與o-DOIMDAA。然而,兩化合物與銅離子生成的錯合物彼此之間卻有著截然不同的螢光量子產率,因此我們藉由NMR來觀測銅離子對兩化合物滴定所造成的訊號的位移與強度的變化,並以此判斷兩錯合物結構上的差異。在後續的光譜分析中,我們發現兩化合物皆具有雙螢光現象,尤其o-DOIMAA在非質子溶劑具有雙螢光現象,但在質子溶劑中卻只能看到單一的螢光峰,所以我們也進一步地對化合物進行酸鹼滴定,並且同樣藉由酸化或中和特定部位所造成的UV和螢光圖譜上的訊號波長以及強度的改變來證實當中的光物理機構。並在最後結合前述的實驗結果對兩化合物在常溫下的熱異構化速率進行探討。
In this experiment, the properties of EDTA were combined to green fluorescent protein, and two polydentate ligand, o-DOIMAA and o-DOIMDAA, which are very selective for copper ions, were synthesized. However, the complexes formed by the two compounds with the copper ions have completely different fluorescence quantum yields. Therefore, we monitored the titration of the two compounds by copper ions with NMR, according to the change of the chemical shift and intensity of the signal to assume the difference of structural between the two complexes. In the subsequent spectral analysis, we found that the two compounds have dual fluorescence, especially o-DOIMAA has dual fluorescence in the aprotic solvent, but only one fluorescent peak in the protic solvent. So we also used acid-base titration to test that two compounds and monitor them with UV and fluorescent spectra, observing the change of absorbance and fluorescence by intensity and wavelength to confirm the truth of photophysical mechanism behind that phenomenon.
