半個多世紀以來,植物細胞的全能性 (totipotency) ,已陸續地從許不同物種利用組織培養的技術得以確認。近年來,隨著動物幹細胞領域研究與醫療、美容業的蓬勃發展,以及草本萃取物大量商品化的廣泛應用,吾輩也開始思考:「在植物細胞具有強大全能分化性的同時,特定物種是否也俱備了足以影響特定動物組織(或細胞)的再生能力?」所以,我們嘗試選用由多種台灣蝴蝶蘭 (Phalaenopsis spp. hybrid) 胚胎所製備之蘭胚素,進而分析其對於不同動物細胞之基本生理性質影響。
實驗初期採用成大蘭花研究中心所提供的四種蘭胚素H0、A2、H3以及H4,以DPPH檢測其抗氧化能力。由結果顯示,H0抗氧化效果最佳,後遂以肌原母細胞 (C2C12) 、 纖維母細胞 (NIH-3T3) 以及角質細胞 (XB2) 檢測MTT生物活性測試、細胞增生能力測試、細胞內毒性測試。並利用黑色素細胞 (B16-F10) 檢測蘭胚素抑制黑色素生成之能力。
Half century ago, the totipotency of the plant cells has been verified by plant tissue culture on many species one after another. In recent years, coming along with the broad application of animal stem cells on medicine, cosmetology and plant extract merchandising; we start to ponder a question, “Whether some specific plant species possess the ability to influence the regeneration of animal tissue or cells when the plant cells acquire the great totipotency.” Hence, we used orchid embryonin from various kinds of Moth Orchid (Phalaenopsis sub.) to analyze its influence on basic physical properties of different animal cells.
At first, the experiment was focused on the four kinds of orchid embryonin: H0, A2, H3 and H4 by NCKU Orchid Research Center to examine the antioxidant capacity through DPPH. This research discovered that the antioxidant effect of H0 was significant. Hence, this research examined MTT assay, proliferation assay, and cytotoxicity assay on C2C12 (myoblast), NIH-3T3 (fibroblast) and XB2 (keratinocytes) through embryonin treatment. Besides, we used B16-F10 (melanocyte) to test its ability of reducing the growth of melanin.
