在MA-1的全合成中,合成出一系列苯環上具有不同取代基之類似物,並將所合成之化合物進一步進行藥理活性測試,其中1-(3,4-dihydroxyphenyl)-4-(4-hydroxy-3-methoxyphenyl)-2,3-dimethylbutane-1,4-dione(25)不論在OGD test模組或glutamate test模組都有不亞於MA-1之活性,因此,具有進一步研究的價值。
在phenanthraquinones的合成研究中,利用於密封管中加入三氯醋酸當催化劑的反應條件,進行Diels-Alder反應,成功將產率提高約10~15%,並利用此方法共合成出25個phenanthraquinones及3個1,4,5,8-phenanthrenediquinones,其中 moniliformediquinone(82)的合成步驟亦較以往文獻中的方法簡單化。由此28個化合物之活性測試結果顯示出其具有結構-活性關係,其中3,6-dimethoxyphenanthrene-1,4,5,8-tetraone(84)的細胞毒殺活性半數有效濃度EC50值範圍介於0.13~0.32 ng/mL,顯示其活性表現相當優異,因此值得進行更進一步的研究。
在將抗發炎藥物及抗癌藥物結合的合成研究中,成功利用醯氯方法及DCC將兩種藥物結合成同時具有抗癌及抗發炎效果之藥物,此實驗共合成出18個化合物,目前仍委託活性測試中。
On the study of total synthesis of analogues of cinnamophilin, we succeed synthesis a series of different substituted groups on benzene ring of lignan. 1-(3,4-dihydroxyphenyl)-4-(4-hydroxy-3-methoxyphenyl)-2,3-dimethylbutane-1,4-dione(25)display significant activities on OGD test and glutamate test, and worth to do further study.
On the study of total synthesis of analogues of phenanthraquinones, we succeed improved the yield about 10~15% by using seal tube and trichloroacetic acid as catalyst. We synthesized twenty five phenanthraquinones and three 1,4,5,8-phenanthrenediquinones by using a concise pathway. The bioactivity displayed structure activity relationship. 3,6-dimethoxyphenanthrene-1,4,5,8-tetraone(84)showed significant activities on cytotoxicity, and the EC50 was about 0.13~0.32 ng/mL.
We design a series of drugs to treat inflammatory and cancer on the same time by using acyl chloride or DCC as catalyst. Finally, synthesized 18 compounds to test it’s bioactivity.
