透明質酸常被利用於組織工程,作為傷口癒合的一種生物醫學材料。但是,由於透明質酸大量存在於人體中,包括在幹細胞微環境中。因此,推測其用途不僅只有當作生物醫學材料,應該還有未被確認的重要功能,其中之一則是維持幹細胞的特性。先前的研究發現透明質酸介面能延長老鼠間葉幹細胞的壽命,而且減少其老化相關之基因表現。我的研究是為了探討透明質酸是否也能維持人類的間葉幹細胞之特性。結果發現透明質酸介面能維持間葉幹細胞的特性:降低細胞型態改變、間葉幹細胞表面抗原表現、骨細胞分化能力、可恢復細胞週期之休眠狀態、端粒酶活性、以及細胞移動能力。特別是維持細胞在一種休眠的狀態,只要把細胞轉移到一般,不含透明質酸介面,細胞則恢復細胞週期,繼續生長。如果細胞已經進入老化,細胞是不會繼續生長的。除此以外,透過高通量分析基因以及蛋白質表現,歸類出了跟透明質酸介面相關的基因以及蛋白質。
The value of mesenchymal stem cells (MSCs) is attributed to the niche from where they originate. Hyaluronan (HA) is a crucial extracellular matrix (ECM) found in the stem cell niche. Previous study reported that HA prevented mouse MSCs from cellular aging with reduced proliferation rate. The HA previously reported to be anti-cellular aging had molecular weight of 1470 kDa. The condition given was the culture surface coated with HA. In this report, the HA was coated at concentration 30 microgram/cm2 (C30), and the condition without HA was termed tissue culture surface (TCS). Initially, the slow proliferation effect of HA was repeatedly found in the human placental-derived MSCs (PDMSCs). As a result, the period of HA treatment was extended. In the young MSCs, cells were consecutively treated for 4 passages (YC30); in the old MSCs, cells were consecutively treated for at least 16 passages (OC30). The OC30 retained MSCs biomarkers expression, osteogenesis differentiation potential, replicative ability, higher telomerase activity even after having transferred to the TCS condition (OC30-->T1) given the identical doubling time as TCS only. This study has compiled the protein and mRNA level of genes associated with proliferation, cell adhesion, cytoskeletal, and HA in the OTCS compared to OC30. In light of this promising finding of HA, the applications of HA is therefore widened, in particular, the foundation of cell biology research related to maintenance of MSCs by HA
