  • Lin 與Shih(2001)針對癌症用藥第一期臨床試驗,傳統A+B 設計(traditional algorithm-based designs),提出試驗期望受試者人數以及每個劑量水準被選為最大耐受劑量的機率等指標,用以瞭解不同A+B 設計的特性,本論文提出允許藥物劑量所對應毒性反應之變異下,修改Lin 與Shih 所提之指標並推導其餘亦可幫助研究人員挑選試驗的指標。

    Lin and Shih (2001) proposed some statistical indexes based on the traditionalalgorithm-based designs for phase I cancer clinical trials. The most interesting indexes are the expected number of patients treated in a trial and the probability of a dose being chosen as the maximum tolerated dose (MTD). These statistical indexes can help the clinical investigators to understand the feature of different A+B designs. In this thesis, we modify their statistical indexes with the variability of toxicity rate under each dose levels. We also derive some other useful statistical indexes which may help trial researchers to choose the suitable design among A+B designs.
