Vibrio anguillarum (鰻弧菌),為革蘭氏陰性菌。是水產養殖漁業中常見的病原菌。V. anguillarum的疫苗已經被發展出來,而且在免疫鮭魚後得到很好的效果。研究報告指出魚體使用弧菌疫苗後,不僅誘發了專一性免疫反應,而且對於它種細菌性疾病也產生了抵抗力。這個結果顯示V. anguillarum可能是一種免疫促進劑。
本實驗發現當石斑魚(E. coioides)經V. anguillarum免疫之後,活化血液中吞噬細胞的氧化殺菌能力(respiratory burst activity),並且非專一性免疫相關基因Mx,在免疫後表現量也明顯上升。這個結果證明V. anguillarum會刺激石斑魚的非專一性免疫反應。
由於V. anguillarum具有這種等同於佐劑的特性,所以,我們發展出弧菌的表現系統,讓弧菌做為次單位疫苗的表現載體。將兩種抗原,綠螢光蛋白(green fluorescent protein)與神經壞死病毒鞘蛋白(nervous necrosis virus coat protein),藉由V. anguillarum表現,分別以注射與口服的方式免疫石斑魚。結果發現以弧菌做為表現載體的次單位疫苗,不論是注射或是口服,免疫效果都優於大腸桿菌次單位疫苗。
Vibrio anguillarum, a Gram-negative bacterium, is a common bacterial opportunist pathogen in marine farm fish. V. anguillarum bacterin vaccine have been developed and successfully used for the immunization of salmonoids, and was found that it could induce specific immune response and also provided cross protection on some other bacterial diseases, indicating that V. anguillarum might be an potential immuno-stimulator.
In this study, we present data indicating that V. anguillarum in grouper(E. coioides) is an immuno-stimulator by demonstrating its bacterin can activate respiratory burst activity of phagocyte, and the expression of an innate immune related Mx protein.
With its capability of an adjuvant activity, subsequently, we have developed a cloning system by using V. anguillarum as an expression host. Two antigens, the GFP and NNV VP antigens, were cloned onto V. anguillarum to construct subunit vaccines, when the subunit vaccine applied either by injection or deliver orally to grouper, it induces stronger specific immune response in grouper compared to that cloned in E.coli.
Our results indicates that V. anguillarum with adjuvant activity makes it a potential vehicle for the development of fish oral subunit vaccine.
