  • 肥胖在已開發國家中是個愈來愈重要的問題,其危機在於造成很多疾病的發生,在歐洲和美國有40%的人遭遇肥胖的問題,研究指出肥胖和愈來愈多疾病的發生有關,例如:高血壓、成人型糖尿病、癌症、心血管疾病。肥胖發生的原因很多,其中一個原因為幹細胞分化為脂肪細胞及脂肪細胞不斷儲存能量變大。有很多治療肥胖的方法:(i) 減少高卡路食物的吸收,(ii) 適當的運動,(iii)減少幹細胞分化為脂肪細胞。

    有研究指出史達汀類的藥物能抑制脂肪前驅細胞分化成脂肪細胞而2-去氧葡萄糖能減少體內葡萄糖代謝。從人類脂肪組織中取得的幹細胞已被證明為間葉世系的幹細胞,能分化成成熟的脂肪細胞。在本篇論文中,我們選擇人類脂肪組織衍生幹細胞作為細胞模式研究史達汀類的藥物及2-去氧葡萄糖的抗脂肪分化影響。初步結果顯示史達汀類的藥物及2-去氧葡萄糖具有抑制幹細胞分化成脂肪細胞之活性。

    Obesity is a more and more important issue in the developed countries. In Europe and the United States, 40% of people are obesity. Obesity is associated with many diseases such as hypertension, diabetes, cancer and cardiovascular diseases. One of the causes for obesity is that the stem cell is induced and differentiated into adipocyte and consequently accumulates excess fat. There are many ways to control obesity; (i) reduce the uptake of high calorie food, (ii) proper exercise, and (iii) block the differentiation of stem cells into adipocytes.
    Statin compounds have been reported block preadipocyte (3T3-L1) differentiated into adipocyte and 2-deoxyglucose has been reported to reduce glucose metabolism in vivo. It has been also demonstrated that stem cell isolated from human adipose tissue is a mesenchymal lineage cell and it can be differentiated into adipocyte. In this report, we chose human adipose tissue derived stem cell as a model system to study the anti-adipogenic effects of statin compounds and 2-deoxy glucose. The results showed that both statin compounds and 2-deoxy glucose have anti-adipogenic activities.
