本研究是使用蛋白質質譜方法來研究寒冷效應(chilling effect)對黑暗中生長的稻米芽鞘的影響。我們挑選剛發芽的稻米芽鞘140顆置於100ml的血清瓶,加80ml的二次水,在室溫中生長,待長到3天後把整瓶稻米芽鞘放入4℃的環境中,不同的時間(0、12hr、1天、6天)。於4℃下,稻米芽鞘的長度仍然繼續拉長,但與在原生長溫度(30℃)下的情況相比較,生長速度確實是緩慢了許多,我們發現在稻米芽鞘在此條件下生長6天的平均存活率仍達90﹪以上。實驗中我們以丙酮沉澱的方法,萃取不同條件下的稻米芽鞘蛋白質,並利用二維電泳分析的方法予以比較,結果顯示出有二個分子量約14KD新的蛋白質,在低溫環境下給合成出來。LC/MS技術將更進一步用來分析鑑定此新的蛋白質點。本研究的結果也暗示著寒冷效應確實會造成稻米發芽時的蛋白質表現之改變。
This study is to use proteomic method to investigate the chilling effect on the protein express for rice coleoptiles. Just germinated rice for about 140 grains were pooled in a 100 ml bottle containing about 80 ml of water and incubated at room temperature for 3 days, which were then shifted to a 4℃ cold room to incubate for various stages (12 hours, 1 day, and 6 days). Coleoptile elongation was observed, but with a slower rate compared with those incubated at room temperature. The average survival rate under chilling condition was more than 90﹪after 6 day of incubation. The total coleoptile proteins extracted from various incubating stages were examined and compared with those incubated at room temperature by using 2-D gel electrophoresis. The extracted protein solution was first treated with acetone to precipitate the proteins out. The precipitated proteins was re-dissolved and then subjected to 2-D gel electrophoretic analysis. It was found two additional protein spots with molecular weight around 14KD were observed for coleoptile incubated at 4℃ for various time. The protein spots will be further identified by using LC/MS spectroscopic technique. These results indicate that chilling cause the physiological change of rice seedling in terms of proteomic view.
