In the early stage of the bladder cancer, target detection of bladder cancer is very important for effective diagnosis and treatment. Until now, cystoscopy and urine cytology are still the mainly detection method for bladder cancer. However, the low sensitivity, invasive detection and expensive price make people don’t want to diagnosis. Surface enhanced Raman scattering (SERS) which has high sensitivity, high identify, noninvasive detection and point of acre can resolve these problems. In our research, we make the SERS substrate through the nanoimprint and photoreduction then apply to the SERS-based immunoassay to detect the Complement Factor H-related protein (CFH), the detection target of bladder cancer, which is approved by U.S. Food and Drug Administration (FDA). In molecule measurement, the detection limit of rhodamine 6G (R6G) and malachite green (MG) is 10-8 M and 10-10 M, respectively. The concentration of MG even lower than the standard of European Union (5.48 x 10-9 M). The uniformity and reproducibility of SERS substrate are great, the relative standard deviation (RSD) is 8.79% and 8.93%, respectively. The CFH test only detect from10-5 g/ml to 10-7 g/ml and success once. The SERS substrate we make not only simple and low cost but also has high uniformity and reproducibility. We provide the more comfortable, more convenient, and cheaper detection method for the bladder cancer patient.
