膠原蛋白為生物體內重要的組成蛋白之一,並被廣泛應用於食品、化妝品、生醫材料等。目前定量總膠原蛋白含量的方法,以傳統羥脯胺酸化學定量分析法為主,過程繁雜耗時且無法確認所有之羥脯胺酸均源自於膠原蛋白。本研究以毛細管電泳儀建立更靈敏、快速、簡便且準確的膠原蛋白定量方法。本論文建立以毛細管電泳分析儀定量膠原蛋白的方法,並確認分析方法的專一性及再現性,此外動物各部位的組織分別以傳統羥脯胺酸化學分析法定量、毛細管電泳儀定量羥脯胺酸含量法定量、毛細管電泳儀直接定量膠原蛋白含量法定量,藉以比較各種定量分析方法之可行性與精確性。由於以上方法之實驗結果均在誤差範圍之內,顯示以毛細管電泳儀定量組織中膠原蛋白的方法確實可行,與至少需要7天的傳統羥脯胺酸化學分析方法比較,我們建立的以毛細管電泳分析儀定量膠原蛋白的方法確實可以更快速的得到正確的定量結果。
Collagen is an important component protein in organism. It has been widely applied in the industries of foods, cosmetics, biomaterials, etc. Its quantification is mainly based on the hydroxyproline assay of samples by chemical chromatic analysis. Since the method is somewhat tedious and non-specific to non-vertebrate samples, the current study focused on establishing an easy, sensitive, and accurate method for the quantification of collagen by using capillary electrophoresis. The specificity and reproducibility were first characterized after identifying the optimal assay system for collagen. Various animal tissues were analyzed by the traditional hydroxyproline assay, hydroxyproline quantification by capillary electrophoresis, and direct collagen quantification by capillary electrophoresis. Our results demonstrated that no significant difference appeared on the data measured by the above variant methods. This indicates that the quantification method developed by using capillary electrophoresis is feasible and accurate. One of the advantages is that our method vastly reduced the timing for waiting and working from about 7 days to less than 3 days which include the time for sample processing.
