As a product developer in TFT-LCD manufacturing, TMAH is highly toxic and refractory and suspected to be an inhibitor in nitrogen removal. Biological processes are currently the most cost-effective treatment method for treating various kinds of wastewater. This study focused on the microbial population dynamics and potential TMAH-degrading methylotrophic bacteria. An aerobic SBR was set to cultivate the TMAH-degrading and nitrifying microorganisms. Sequencing and quantitative PCR were then applied to investigate the microbial population during the TMAH-degrading process. The results showed complete degradation of a high level of TMAH, followed by its nitrification. The sequencing study identified successful enrichments of Hyphomicrobium, Nitrosomonas and Nitrospira in the aerobic SBR fed with high levels of TMAH. The cloning also successfully developed the standard plasmid for tmm gene. The qPCR revealed an initial inhibition of AOB and Thiobacillus by TMAH. The role of Hyphomicrobium and Thiobacillus as methylotrophic bacteria were also confirmed. Hyphomicrobium might play a role in the degradation of TMAH while Thiobacillus plays a role in the degradation of intermediate TMAH species.
