  • sparc(secreted protein acidic and rich in cystein /osteonectin )基因所轉譯出的SPARC蛋白屬於一種細胞間質蛋白,具有調節細胞與細胞外基質黏附性及細胞訊息調控的功能,主要分布在骨頭與細胞間質中,其所掌控的生理現象早至胚胎發育過程,細胞的分化、組織的鈣化生成、骨骼發育、型態及器官發育有極大影響;在哺乳類的研究之中也發現SPARC基因具有調節脂肪形成的能力。本論文從活體內及活體外的層面來對SPARC蛋白進行功能進行探討;在活體外:從細胞實驗上驗證石斑魚重組SPARC蛋白在體外的生物性功能與其他物種研究相似,建立一套便於觀察型態變化系統,而由真核細胞所表現的SPARC重組蛋白具有與內生性蛋白更相似的修飾作用及分泌作用;而活體內實驗,藉由觀察在健康、神經壞死病毒感染石斑魚上和不同發育時期以及各組織內sparc基因表現量,推測SPARC在生理功能上所扮演的角色;另外在降低魚體內內生性SPARC蛋白的動物實驗上證明SPARC能抑制脂肪新生,並且與myostatin具有相關聯性。

    SPARC (secreted protein acidic and rich in cystein /osteonectin) protein is a matricelluar protein which is mostly expressed in bone and ECM, could modulate the interaction between cells and extracellular matrix and regulate signal transduction. The function of SPARC protein is involved in the development of ossified and mineralized tissues, tissue morphogenesis and cellular differentiation. Besides, SPARC protein may also regulate fat production. Here, we will study the functions of SPARC protein in vitro and in vivo. First, in vitro, we prove SPARC protein of grouper has the Biological function same as other species and we established a system to easy observation of morphological change. Second, in vivo, we prove SPARC protein can inhibit adipogenesis and it is associated with myostatin. On the other hand, we discussed the physiological important by observing the expression level during embryo development, the fish of healthy and infected NNV, and different tissues.
